Start winuae as administrator?

Probably an NTFS file permissions problem, you are not allowed to write in there.

"Program Files" is write-protected by default since Vista.

On Vista and 7 you should only put winuae.exe into C:\Program Files\WinUAE. Everything else shoud go into C:\Users\Public\Amiga Files. WinUAE will automatically find its stuff there.

When moving from XP to Vista or 7 you might have to change access permissions and/or ownership of the directory and might be even every single file.
