Once the copper is displayed, would any changes be immediate or would I need to recall the copper?

It's safe to change the copper list on the fly, but just make sure you're changing the list where the copper isn't reading. A good place would be at the end of a visible frame: Wait until the end of the screen, then change the color values for the copper bars in the copper list.
