The two main ways are the MIDI one and the tracker one. What to use depends on your goal/preferences.

For MIDI you need a MIDI-interface where you can connect MIDI instruments and a MIDI-sequencer program like BarsnPipes for control/arrangement. I don`t know it it works with WinUAE or if it makes sense.

Then there are 4 channels (Octamed, Protracker, ...) and multi channels trackers (OctamedSoundStudio, DigiboosterPro, HivelyTracker, ...) what are sequencer programs. They usually use samples (recorded instruments, sounds, voices, ...) you can load and arrange.
